The collection of these data utilizes a Burke-o-Lator (BoL) pCO2/TCO2 analyzer and began August 30, 2023. The Burke-o-Lator (BoL) pCO2/TCO2 analyzer measures carbon dioxide partial pressure (pCO2) and total dissolved inorganic carbon (TCO2) from a flow-through seawater stream and from seawater collected in discrete samples. The BoL is paired with a SBE 45 Thermosalinograph to measure seawater temperature and salinity. These data provide valuable information on patterns in marine carbonate chemistry and ocean acidification within Barkley Sound waters being supplied to the Bamfield Marine Sciences Centre and the Nova Harvest Ltd. shellfish hatchery. The service route of the Columbia included the ports of Sitka, Juneau, Haines, Skagway, Wrangell, Petersburg, and Ketchikan in southeast Alaska, and between Ketchikan and Bellingham, WA. The vessel sailed south from Ketchikan every Wednesday, arrived in Bellingham on Friday, returned to Ketchikan on Sunday, and transited between southeast Alaska terminals between Sunday and Wednesday. Measurements of temperature, salinity, dissolved oxygen content, and CO2 partial pressure were made every 2 mins on seawater drawn into the vessel from a ~2 m intake. Seawater was delivered from the intake to analytical equipment on the car deck using a AMT 1/2 HP pump (4295-98). Temperature and salinity data were collected using a Sea-Bird SBE 45 MicroTSG Thermosalinograph, and intake temperature measured using a SBE 38 Digital Oceanographic Thermometer. Dissolved oxygen content was determined using an Aanderaa 4330F that had undergone multipoint calibration at the factory in Norway. CO2 measurements were made using a General Oceanics 8050 (GO8050) pCO2 Measuring System equipped with a LI-COR LI840A non-dispersive infrared detector. All data streams, including GPS information from an antenna and atmospheric pressure from a Vaisala barometer positioned on the foredeck, were captured by the GO8050 control computer. Atmospheric CO2 measurements were also made using the GO8050 on unaltered marine air drawn to the system through an intake located on the foredeck. The CO2 measurement scheme involved the analysis of four gas standards of known CO2 content (150 ppm, 349 ppm, 449 ppm, and 850 ppm; Praxair Distributions Inc), 12 analyses atmospheric CO2, and 240 seawater CO2 measurements in a 8.5 hour period. The atmospheric and seawater CO2 analyses were run in a sequence of 3 atmospheric measurements and 60 seawater measurements that was repeated 4 times between standardization. This project was supported by the Tula Foundation and Alaska Ocean Observing System, and was a collaborative effort between the Hakai Institute, University of Washington Joint Institute for the Study of the Atmosphere and Ocean/NOAA Pacific Marine Environmental Laboratory, State of Alaska Department of Transportation, University of Alaska Southeast Alaska Coastal Rainforest Center, and the Alaska Ocean Observing System. Sample water (250 ml) was filtered through: 1) a single glass fiber filter (GF/F, nominal pore size 0.7 um) to comprise a bulk measure of phytoplankton chlorophyll and phaeopigment concentrations and 2) a stack of filters (GF/F, 3 um, and 20 um polycarbonate filters) to estimate chlorophyll and phaeopigment concentrations from pico, nano, and micro-phytoplankton, respectively. Filters were extracted for 24 hours in 10 ml of 90% acetone and analyzed on Turner Designs Trilogy laboratory fluorometers using the acidification module and following the method of Holm Hansen et al. Hakai Bulk and Size-Fractionated Chlorophyll and Phaeopigment Concentrations Collected by Niskin Bottle, Provisional - The utilized acidification method is sensitive to interference by high concentrations of chlorophyll c and b. High chlorophyll c concentrations can cause a slight overestimation of chlorophyll a and an underestimation of phaeopigments, which can result in negative phaeopigment concentrations. High chlorophyll b concentrations can result in underestimations of chlorophyll a and overestimations of phaeopigments. When chlorophyll b is high relative to chlorophyll a, chlorophyll a can be underestimated by as much as 2.5 fold. For more details see Welschmeyer et al. (1994). These two types of interference likely had an influence on the published chlorophyll and phaeopigment dataset. Notably, comparison with HPLC has shown that stations in the Quadra Island region often experience conditions where chlorophyll b is present in high concentrations and is high relative to chlorophyll a. The acidification method used to analyze the Hakai Institute dataset has been shown to highly overestimate phaeopigment concentrations based on comparison to concentrations determined by HPLC (Trees et al., 2003). Care should be used when interpreting the published phaeopigment concentrations. Prior to 2021, samples that exceeded the manufacturer specified instrument sensitivity range were not diluted to bring their concentrations within the specified range. Samples run above the instrument sensitivity range may have underestimated chlorophyll concentrations due to the blockage of light passing through the sample and a corresponding reduction of fluorescence output (see Turner Trilogy user manual for complete description). Comparison with matching HPLC derived chlorophyll a suggests that the affected samples may be useable, but further analysis is required. This provisional dataset includes these data. Prior to 2018, the calibrations of the Hakai Institute Turner Trilogy benchtop fluorometers showed non-linear variability in the calibration slopes and acid ratios that were applied to the data. This variability suggests error due to analytical methods rather than instrument drift. As a result, the chlorophyll and phaeopigment concentrations of the data where these calibrations were applied may have error as high as +/- 12 % from this source alone. These data have been included in this provisional dataset. Prior to 2018, QA/QC analysis suggests potential influence by acid contamination on the chlorophyll and phaeopigment concentrations. To limit this influence, disposable cuvettes were used for samples analyzed following 2018 and additional methodological controls were put in place to limit acid contamination. Care has been taken to flag contaminated data, but data with less obvious contamination effects may have not been captured by QA/QC and may be present in this provisional dataset. This provisional dataset includes all data flagged as "suspicious" (show suspect trends, but may not be incorrect), but excludes those flagged as "discard" (QA/QC analyses showed they erroneous). + ##Limitations: +The utilized acidification method is sensitive to interference by high concentrations of chlorophyll c and b. High chlorophyll c concentrations can cause a slight overestimation of chlorophyll a and an underestimation of phaeopigments: negative phaeopigment concentrations can occur during bloom events with high chlorophyll c and healthy phytoplankton. (1994). Justin Del Bel Belluz - www.hakai.org - This dataset is comprised of Hakai Institute chlorophyll and phaeopigment concentrations collected from 2017 to present at stations within the Calvert Island, Johnstone Strait, and the Quadra Island study regions. Samples for these data were collected in the field using Niskin bottles. (1965). Data have been quality controlled by manual inspection which included a comparison of bulk concentrations against the sum of the size-fractionated filter concentrations. Where available, further comparisons were done with other measures of chlorophyll concentrations including in situ CTD chlorophyll fluorescence and high-performance liquid chromatography (HPLC) data. Data were collected by the Hakai Institute Oceanography and Nearshore programs. + Del Bel Belluz, Justin + https://www.hakai.org/ + This dataset is comprised of Hakai Institute chlorophyll and phaeopigment concentrations collected from 2017 to present at stations within the Calvert Island, Johnstone Strait, and the Quadra Island study regions. Hakai Bulk and Size-Fractionated Chlorophyll and Phaeopigment Concentrations Collected by Niskin Bottle, Research The utilized acidification method is sensitive to interference by high concentrations of chlorophyll c and b. As a result these samples were removed and the dataset lacks many concentrations from phytoplankton bloom events. Comparison with matching HPLC derived chlorophyll a suggests that the affected samples may be useable, but further analysis is required.
